Product Description: Isocitrate dehydrogenase (IDH) catalyzes the oxidative decarboxylation of isocitrate to alpha-ketoglutarate using either
NAD+ or NADP+ as the electron acceptor. NAD-dependent IDH (or IDH3) is a highly regulated enzyme of the TCA cycle and is a multisubunit
enzyme localized in the mitochondrial matrix. IDH1 and IDH2, on the other hand, are dependent on NADP+, and catalyze reversible reactions. IDH1
is primarily found in peroxisomes and cytosol and IDH2 is located in the mitochondria. Since IDH1 and IDH2 mutations are prevalent in several
types of malignancies, IDH targeting therapies have emerged as potential cancer treatments. Indeed, several clinical trials with various IDH
inhibitors are ongoing. Of note, IDH mutations are found to lead to increased conversion of alpha-ketoglutarate to 2-hydroxyglutarate, which
increases cancer cell proliferation and metastasis in part through DNA methylation and/or activation of VEGF receptor. Given that IDH-mutated
cells utilize alpha-ketoglutarate to produce 2-hydroxyglutarate and depend on glutaminolysis as a source of alpha-ketoglutarate, use of
glutaminase inhibitors is a potential approach to suppress IDH mutant cancers. The non-radioactive colorimetric IDH1/2 assay is based on the
reduction of the tetrazolium salt INT in a NADP-coupled enzymatic reaction to formazan, which is water-soluble and exhibits an absorption maximum
at 492 nm. The intensity of the red color formed is increased in the presence of increased IDH activity. Each kit is sufficient for 100 wells of 96-well
plates. The IDH assay solution should be stored in aliquots at -80ºC, and will remain stable indefinitely at -70ºC.
