Members of the caspase family of cysteine proteases are critically involved in coordinating the events occurring during apoptosis. Caspases are activated in response to diverse death signals and ultimately demolish the cell through limited proteolysis of more than 400 protein substrates. Caspases are initially produced as inactive monomeric procaspases that require dimerization and often cleavage for activation. In humans, caspase-8, caspase-9, and caspase-10 play upstream roles, i.e., initiator caspases, by coupling cell death to the downstream effector or executioner caspases (caspase-3, caspase-6, and caspase-7). Various apoptotic pathways exist (extrinsic vs. intrinsic) that can be distinguished by the adapter proteins and initiator caspases involved. While the initiator caspases primarily cleave their own precursors and the downstream effector caspases, the bulk of the proteolysis that takes place during apoptosis is carried out by the executioner caspases. The Caspase 3 activity assay kit is based on proteolytic hydrolysis of the chromogenic peptide substrate Asp-Met-Gln-Asp-p-nitroanilide. Cleavage of pNA from the substrate increases absorbance at 405 nm (extinction coefficient= 9.5 mM-1cm-1), allowing for sensitive and quantitative assay of Caspase 3 activity present in tissue/cell lysates. Kit components are stable for at least 1 year if stored and handled properly.
