Adenylate kinase (ADK or myokinase) catalyzes the reversible transfer of the terminal phosphate group between ATP and AMP (ATP + AMP <=> 2 ADP), i.e., the enzyme relays adenine nucleotide signals to metabolic sensors. This ubiquitous enzyme is thus involved in energy metabolism and nucleotide synthesis. Two ADK isozymes have been identified in mammalian cells: ADK1 is cytosolic and ADK2 is mitochondrial. ADK1 deficiency in the erythrocyte is associated with hemolytic anemia. ADK1 knockout disrupts the balance between ATP consumption and ATP synthesis and energy homeostasis in the post-ischemic heart, precipitating myocardial ischemia-reperfusion injury. ADK2 deficiency in humans causes hematopoietic defects. If abundant ADP is present as the substrate, the ADK-catalyzed reaction is driven toward formation of ATP, which provides a linear production of ATP and can be used for measuring ADK activity in serum/plasma and crude extracts using the luciferase/luciferin system. Luciferase-based detection of ATP produced by ADK reaction allows fast, sensitive and quantitative assay of ADK activity. Measurement of ATP requires a luminometer or scintillation counter. Kit components are stable for many years if stored and handled properly.
