Aspartate aminotransferase (AST) is a member of the transaminase family of enzymes. The enzyme is previously referred to as glutamate oxaloacetate transaminase (GOT), which indicates its biological function in intermediary metabolism of glucose and protein through catalyzing the transfer of an amino group between aspartate and glutamate. AST is found in many tissues throughout the body, including the liver, heart, muscle, brain, and kidney, and therefore AST measurement along with ALT measurement is routinely performed in clinical diagnostics. While serum levels of AST are elevated in most types of liver disease, increased serum AST may also be associated with myocardial infarction, renal damage, and muscle necrosis. Assessment of the ratio of AST to ALT also provides clinically useful information. The colorimetric AST assay is based on the reduction of the tetrazolium salt INT in a NADH-coupled enzymatic reaction to formazan, which is water-soluble and exhibits an absorption maximum at 492 nm. The intensity of the red color formed is increased in the presence of increased AST activity. The kit is stable for at least one year if handled and stored properly.
