Ketone bodies, like short chain fatty acids, are water-soluble, and include acetoacetate, beta-hydroxybutyrate, and acetone. Liver mitochondrion is the major site of ketogenesis, which occurs in diabetic ketosis and whenever fatty acid levels are elevated in the blood or as a result of a high fat, low-carbohydrate diet, e.g., Atkins diet. Although the brain cannot burn fatty acids, it can use ketone bodies during starvation. Notably, ketone bodies have been successfully used for the treatment of epilepsy, and may be beneficial for patients with neurodegenerative disorders. The enzyme beta-hydroxybutyrate dehydrogenase (BDH), a lipid (PC)-requiring enzyme that is found on the inner surface of the inner mitochondrial membrane, mediates the interconversion between acetoacetate and beta-hydroxybutyrate. While the BDH reaction favors the production of beta-hydroxybutyrate in the liver, the reaction is reversed in the brain and peripheral tissues for ketone body utilization. The non-radioactive colorimetric BDH activity assay is based on the reduction of the tetrazolium salt INT in a NADH-coupled enzymatic reaction to formazan, which is water-soluble and exhibits an absorption maximum at 492 nm. The intensity of the red color formed is increased in the presence of increased BDH activity. The kit is stable for at least one year if handled and stored properly. Each kit is sufficient for 100 wells of 96-well plates. The BDH assay solution should be stored in aliquots at -80ºC, and will remain stable indefinitely at -80ºC.
