Nicotinamide adenine dinucleotide (NAD) is a coenzyme involved in cellular metabolism and energy production. NAD+ levels tend to decline with age, and interventions aimed at boosting NAD+ levels have been explored as potential anti-aging strategies. CD38 is an ectoenzyme possessing a major activity of hydrolyzing NAD+ (NADase activity). In this context, CD38 is implicated in several pathways that may influence the aging process. The NADase enzyme activity of CD38 is involved in the regulation of intracellular calcium levels and cellular signaling pathways. CD38 also possesses the activity of synthesis of ADP-ribose (ADPR) and cyclic ADP-ribose (cADPR) from NAD+. Although CD38 can be found in the cytoplasm and in the membranes of intracellular organelles, the vast majority of CD38 activity is in the plasma membrane, facing outside the cell. CD38 is expressed on the surface of various immune cells, including T cells, B cells, and natural killer (NK) cells, and is strongly linked to tissue inflammation. Its expression levels can be up-regulated in response to inflammatory stimuli. Since NAD+ decline is linked to the development of multiple age-related conditions and diseases, CD38 is a potential diagnostic and therapeutic target. The CD38 Activity Assay Kit is based on the use of NAD+ as substrate, which is converted to NADH through lactate dehydrogenase-mediated catalysis for assay detection. The resultant NADH reduces the tetrazolium salt INT to formazan, which is water-soluble and exhibits an absorption maximum at 492 nm. The assay kit is suitable for pharmaceutical screening of CD38 inhibitors. Kit components are stable for at least one year if stored and handled properly.
