Dipeptidyl peptidase-4 (DPP-4 CD26) is a serine exopeptidase expressed on the surface of most cell types, capable of cleaving X-proline or X-alanine dipeptides from the N-terminus of polypeptides. The DPP-4 and fibroblast activation protein (FAP) have a 70% identity at the amino acid sequence. There are more than 60 identified protein substrates of DPP-4, including chemokines, growth factors, neuropeptides and vasoactive peptides. The pleiotropic effects of DPP-4 regulate glucose metabolism, inflammation, appetite, immune function and extracellular matrix remodeling. Notably, inhibitors of DPP-4 have emerged as a new class of antidiabetic drugs through stabilization of two key glucose-regulating hormones: glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1). In addition, Middle East respiratory syndrome coronavirus (MERS-CoV) is known to use DPP-4 as receptor for cell entry. The DPP-4 activity assay kit is based on proteolytic hydrolysis of the chromogenic dipeptide substrate Gly-Pro-p-nitroanilide hydrochloride (Gly-Pro-pNA) under an optimized condition. Cleavage of the pNA from the dipeptide increases absorbance at 405 nm (extinction coefficient= 9.5/mM-cm) and 410 nm (extinction coefficient= 8.8/mM-cm), allowing for sensitive and quantitative assay of DPP-4 activity present in tissue/cell lysates and biological fluids. Kit components are stable for ~1 year if stored and handled properly.
