The de novo pathway for the synthesis of guanosine monophosphate (GMP) from xanthosine monophosphate (XMP) is mediated by GMP synthase (GMPS), which is one of three amidotransferases involved in de novo purine synthesis. Although there are three possible enzymatic reactions for the generation of GMP, i.e., those catalyzed by GMPS, HGPRT, and guanosine kinase, GMPS is an essential enzyme in many pathogenic organisms, making the enzyme a key drug target. Differences between the human and pathogen GMPS make it a promising target for developing drugs that can selectively inhibit the microbe without harming the host. For example, GMPS is crucial for the pathogenicity of fungi like Candida albicans and Aspergillus fumigatus. Notably, GMPS is overexpressed in many malignant tumors, including lung, ovarian, and head and neck cancers and the enzyme can promote cell invasion and cancer growth. The GMPS inhibitor angustmycin A has been shown to suppress melanoma cell invasion and tumor growth in animal models. The enzyme activity assay kit takes advantage of the glutaminase activity of GMPS and relies on the detection of glutamate, which is coupled to the reduction of the tetrazolium salt INT in a NADH-coupled enzymatic reaction to formazan, which exhibits an absorption maximum at 492 nm.
