The bacterial enzyme beta-galactosidase (LacZ) is a widely used reporter enzyme, the activity of which can be conveniently measured spectrophotometrically (J. Med. Chem. 7:574,1964). The beta-Galactosidase Staining Kit is based on the histochemical substrate 5-bromo-4-chloro-3-indolyl-b-D-galactopyranoside (Xgal) for in situ staining of transgenic tissues or monolayer cells transiently or stably expressing E. coli beta-galactosidase (see LacZ Assay Kit). The cells expressing b-galactosidase stain blue after mild fixation and substrate exposure. The staining kit requires minimal steps for staining the enzyme. This method provides rapid information regarding exogenous expression of beta-galactosidase in animal tissue and transfection efficiency in cell culture. Note that a 4% paraformaldehyde solution is required for cell fixation, but is not included in the kit.
