The non-essential amino acid glutamate is a key molecule in cellular metabolism, serving as an intermediate in transamination and deamination reactions, which allows nitrogen from amino acids to be excreted. Glutamate is also a critical neurotransmitter involved in learning and memory. In brain injury however, glutamate accumulates outside cells, causing excitotoxicity. In the food industry, glutamate is widely used as a flavor enhancer. The side chain carboxylic group has a pKa of 4.1, thus conferring a negative charge at physiological pH. Our non-radioactive colorimetric glutamate assay is based on the reduction of the tetrazolium salt INT in a NADH-coupled enzymatic reaction to formazan, which is water-soluble and exhibits an absorption maximum at 492 nm (detection limit ~20 uM). The intensity of the red color formed is increased in the presence of increased L-glutamate. Please note that tissue, plasma, and serum samples need to be deproteinized prior to assay. The Glutamate Assay Solution is stable for several years if stored and handled properly.
