Nucleoside diphosphate kinase (NDK or NDPK) catalyzes the exchange of phosphate groups between different nucleoside diphosphates with little specificity for the base and the sugar: ATP + NDP <=> ADP + NTP. Thus, nucleotide analogues used in antiviral therapy are also phosphorylated by NDK to their triphosphate forms. In energy metabolism, NDK serves to maintain an equilibrium between the concentrations of different nucleoside triphosphates. For example, GTP produced in the Krebs or TCA cycle is readily converted to ATP through the mediation of NDK, and the enzyme has a major role in the synthesis of nucleoside triphosphates other than ATP. There are two isoforms of NDK in higher eukaryotes: NDK-A and NDK-B, which have similar structure and function, and can combine in any proportion to form functional NDK hexamers. The enzyme activity assay kit can measure NDK activity present in serum/plasma and crude cell/tissue extracts. Luciferase-based detection of ATP produced by NDK reaction allows fast, sensitive and quantitative assay of NDK activity. Measurement of ATP requires a luminometer. Kit components are stable for many years if stored and handled properly.
