Protein Disulfide Isomerase (PDI) Assay Kit

Protein disulfide isomerase (PDI), a prototypic member of a family of ~20 PDI-like proteins, is an ER-resident enzyme and ER marker that catalyzes the formation and cleavage of disulfide bonds during protein folding. This function of PDI includes the oxidation of two cysteines to form a disulfide bridge, the reduction and isomerization of disulfide bonds, and prevention of misfolded protein aggregates. Although PDI has an ER retention signal sequence, it can be secreted from a variety of cells and modulates cell surface protein interaction. Extracellular PDI appears to have a role in thrombosis and vascular inflammation. Although PDI is widely expressed in many tissues, its expression is enriched in the pancreas and liver, and is upregulated during ER stress. Notably, expression of PDI is altered in various cancers, cardiovascular diseases, and neurodegenerative disorders. Our PDI activity assay kit is modified from the documented turbidometric assay with vastly improved sensitivity. As little as 1 mcg of crude cell/tissue can be used for detection of PDI activity. The reduced insulin chains aggregate and the turbidity is monitored at 600-650 nm. The kit is designed to assay PDI activity present in crude cell/tissue lysates. The assay can be adapted to screen for small molecule inhibitors of PDI.
