Quantification of inorganic as well as organic phosphates (such as phospholipids, phosphoproteins, and nucleid acids) is of interest in many biological studies. Phosphatase activity, for instance, can be derived from such an analysis after co-incubation of a phosphatase and a specific phosphopeptide substrate (J. Cell. Biochem. 77:445,2000). Similarly, an assay for phospholipase activity can be performed after hydrolysis of phospholipids by phospholipases. Although various methods can be employed to measure phosphates, non-radioactive colorimetric protocols suitable for high-throughput analysis are the methods of choice in most laboratories. Perhaps the most sensitive colorimetric method for phosphate assay is based on complex formation of phosphomolybdate with the basic dye Malachite Green (Anal. Biochem. 161:45,1987). Our Phosphate Assay Kit (sufficient for 400 assays) is formulated to maximize the sensitivity and stability of the dye-phosphomolybdate reaction. The kit is stable for at least one year if handled and stored properly. The assay, compatible with microplate formats, is simple and can be finished in 15 - 20 min. Reaction product is quantified by absorbance at 600-660 nm.
