Threonine is an essential amino acid with critical nutritional and metabolic function. Two major pathways for the degradation of L-threonine in mammals are those mediated by threonine dehydratase and threonine dehydrogenase (TDH). While the former enzyme converts threonine to ammonium and 2-ketobutyrate, which is eventually oxidized to CO2, TDH forms 2-amino-3-ketobutyrate. 2-amino-3-ketobutyrate is further cleaved to form glycine and acetyl-CoA, or undergoes spontaneous decarboxylation yielding aminoacetone and CO2. TDH is a member of the zinc-containing long-chain alcohol/polyol dehydrogenase family. Threonine catabolism mediated by TDH has recently been recognized as a specific metabolic trait of mouse embryonic stem cells, possessing a potential regulatory role in stem cell programming. Our non-radioactive colorimetric TDH assay is based on the reduction of the tetrazolium salt INT in a NADH-coupled enzymatic reaction to formazan, which is water-soluble and exhibits an absorption maximum at 492 nm. The intensity of the red color formed is increased in the presence of increased TDH activity. The kit is stable for many years under proper storage and handling conditions.
