Tartrate-resistant acid phosphatase (TRAP), encoded by the ACP5 gene, is highly conserved throughout mammalian cells, playing a significant role in processes such as bone mineralization, ECM remodeling, cytokine production, and immune regulation. Enzyme catalysis is mediated through the binding of a phosphate substrate to the Fe2+ in the active site of TRAP. Bone metabolism is coordinated between osteoblast-mediated bone formation and osteoclast-mediated bone resorption. Since TRAP is expressed by osteoclasts, the enzyme and alkaline phosphatase (ALP) are used as markers of osteoclasts and osteoblasts, respectively, and their serum levels serve as a diagnostic for relevant bone diseases. In addition, TRAP is also a lysosomal enzyme. Expression of TRAP can be increased in several other pathological conditions, e.g., hairy cell leukemia, HIV- induced encephalopathy and Gaucher’s disease. The assay kit is based on conversion of p-nitrophenol phosphate to nitrophenol in an acidic buffer. Nitrophenol exhibits an absorption maximum at 405 nm (molar extinction coefficient = 18.75 mM-1cm-1), allowing for sensitive detection of TRAP activity in crude samples. Assay solution is stable for several years if stored and handled properly.
