Xanthine dehydrogenase (XDH), xanthine oxidase (XO), or xanthine oxidoreductase (XOR) is a complex metalloflavoenzyme that contains both iron and molybdenum, catalyzing the oxidation of hypoxanthine to xanthine to uric acid. These enzymes are single-gene products that exist in separate but interconvertible forms. In normal tissue, ~90% of the enzyme is present as the “D form or Dehydrogenase”, which transfers electrons to NAD+ and ~10% is present as the “O form or Oxidase”, which transfers electrons to oxygen to form superoxide and hydrogen peroxide. Human XDH is located predominantly in the liver, small intestine and capillary endothelium. Of note, XDH can be irreversibly converted to XO by proteolysis or reversibly through the oxidation of sulfhydryl groups during tissue injury. The mechanism for the production of reactive oxygen species in ischemic tissue also involves changes in purine metabolism due to reduced ATP production. The non-radioactive colorimetric XDH Assay Kit is based on the reduction of the tetrazolium salt INT in a NADH-coupled enzymatic reaction. The INT reaction product is water-soluble and exhibits an absorption maximum at 492 nm. The intensity of the red color formed is increased in the presence of increased XDH activity. The enzyme assay solution should be kept at -80C, and will remain stable at -80C for at least 1 year.
