Product Description: Alanine aminopeptidase (AAP; aminopeptidase N or CD13) belongs to the group of exopeptidases that split single amino
acids from the N-terminus of the peptide chain. AAP is a zinc-binding membrane glycoenzyme found in many tissues and biological fluids. AAP in
serum largely originates from the liver, and its activity is markedly increased in hepatobiliary disease. Since AAP is released into urine from the
brush border membrane of the renal proximal tubule, damage to the tubules caused by nephrotoxic substances may lead to urinary excretion of
this enzyme. In the small intestine, AAP plays a role in the final digestion of peptides generated from hydrolysis of proteins by gastric and
pancreatic proteases. Notably, the cell surface AAP is a major cell entry receptor for certain strains of coronaviruses, e.g., alpha-genus PRCV.
The AAP activity assay kit is based on proteolytic hydrolysis of the chromogenic substrate L-Ala-p-nitroanilide (Clin Chem 26:1251, 1980).
Cleavage of pNA from the substrate increases absorbance at 405 nm (extinction coefficient= 9.5 mM-1cm-1) and 410 nm (extinction coefficient=
8.8 mM-1cm-1), allowing for sensitive and quantitative assay of AAP activity present in tissue/cell lysates and biological fluids. Kit components are
stable for at least 1 year if stored and handled properly.
