Copper is an essential trace element in human nutrition and plays several critical roles in the body due to its involvement in enzymatic reactions, cellular respiration, iron metabolism, and antioxidant defense. Hemocyanin, a copper-containing protein, is involved in oxygen transport in invertebrates. Mammalian enzymes that require copper as cofactors include cytochrome c oxidase, superoxide dismutase, lysyl oxidase, tyrosinase, and ceruloplasmin. Ceruloplasmin is a copper-containing protein involved in iron oxidation, which is necessary for iron transport. Copper deficiency can lead to anemia due to impaired iron mobilization. Menkes disease, a genetic disorder, impairs copper absorption or utilization. Wilson’s disease is another genetic disorder where copper accumulates in tissues due to a defect in copper excretion, leading to liver and brain damage. Other prominent roles of copper are in the synthesis of neurotransmitters (norepinephrine) and production of the black pigment melanin. The recommended daily intake of copper for adults is approximately 0.9 mg per day. Our Copper Assay Kit is based on reduction of cupric ion to cuprous ion under an alkaline pH and the cuprous ion then reacts with bicinchoninic acid to form a purple-colored complex that absorbs light at 540-562 nm. The intensity of the color is proportional to the amount of copper. The assay detection limit is 1-10 uM.
