Iron is an essential trace mineral that is involved in numerous physiological and cellular processes. As a key component of hemoglobin and myoglobin, iron facilitates oxygen transport between tissues. Iron-containing enzymes and cytochromes play vital roles in ATP production, metabolism, DNA synthesis, and immune function. Iron deficiency is the most common nutritional insufficiency globally and can lead to anemia, cognitive impairment, and immune disorder. On the other hand, iron overload, e.g., hemochromatosis and repeated blood transfusions, can cause tissue damages due to elevated oxidative stress, and iron accumulation in the brain has been linked to neurodegenerative diseases. Our Iron Assay Kit is based on a modified Ferene iron-binding method and is suitable for analysis of iron present in cells and biological fluids such as serum/plasma, saliva, and urine. The ferene iron probe has an absorbance max at 593-595 nm when complexed with ferrous iron (Fe2+). In the assay, an acidic buffer is used to release iron from iron carrier proteins and a reducing agent (ascorbic acid) is present to convert ferric iron (Fe3+) to ferrous iron (Fe2+). In addition, a copper chelator is added to avoid interference from copper ion. The kit can also measure Fe3+ by skipping the iron reduction step. The kit measures iron in the linear range of 10-200 μM.
