Glutathione (GSH) is a tripeptide, gamma-L-glutamyl-L-cysteinyl-glycine, involved in diverse cellular function such as antioxidant defense, metabolic reaction, signal transduction, protein-S-glutathionylation, cell growth and survival. It is the most important hydrophilic antioxidant, guarding cells against exogenous and endogenous toxins, including reactive oxygen and nitrogen species. Decreased levels of GSH are implicated in the etiology of many human diseases, contributing to oxidative stress associated with aging, inflammation, infection and neurodegeneration. In healthy state, more than 90% of the total glutathione pool exists in the reduced form and less than 10% in the oxidized form (GSSG). Although GSH is known to participate in both conjugation and reduction reactions mediated by glutathione S-transferase (GST), it is also capable of engaging in non-enzymatic conjugation with some chemicals. The GSH Assay Kit is based on a spectrophotometric assay (Habig et al., PNAS 71:3879-3882, 1974), which employs 1-chloro-2,4-dinitrobenzene (CDNB) as a substrate for GST-mediated GSH conjugation, causing an increase in the absorbance at 340 nm. The assay requires a 0.2-ml Quartz cuvette or UV-transparent 96-well plate for measuring increased O.D.340 nm. Each assay kit is for 100 assays.
