Creatine (N-aminoiminomethyl-N-methyl glycine) is synthesized from arginine and glycine by arginine glycine amidinotransferase (AGAT) to guanidinoacetate in the body, which is then methylated with S-adenosyl methionine to form the high-energy metabolite. Creatine and phosphocreatine are critically involved in providing cellular energy through creatine kinase (CK), which catalyzes creatine conversion to phosphocreatine. Notably, spontaneous formation of creatinine occurs during the CK reaction and creatinine is primarily removed by the kidneys. If the kidney function is impaired, blood creatinine levels will rise, making blood and urine creatinine concentrations useful for calculation of the glomerular filtration rate (GFR). Serum creatinine is the most used indicator of renal function. Since all the creatinine produced in the body is normally excreted in the urine, analysis of urine creatinine is included in standard urine drug tests because an expected creatinine concentration indicates that the test urine sample is undiluted. The Creatinine Assay Kit is based on the Jaffe reaction, a chromagenic method used in clinical chemistry to measure blood and urine creatinine. The assay can be performed using a 96-well plate and measurement is based on light absorbance at 490-510 nm. The kit is stable for at least one year if stored and handled properly.
