Phosphoglucose isomerase (PGI as known as GPI and PHI) mediates the interconversion between glucose-6-phosphate (G6P) and fructose-6-phosphate (F6P) in glycolysis and gluconeogenesis. Since the enzymatic step is reversible, the direction of the reaction is determined in part by G6P and F6P concentrations. In addition to its role in glucose metabolism, PGI also functions extracellularly. PGI secreted by cancer cells is known as autocrine motility factor (AMF), which through AMF receptor promotes cancer metastasis. PGI has also been described as a neurotrophic factor and maturation factor in its diverse extracellular function. Clinically, PGI deficiency can lead to hemolytic anemia due to impaired glycolysis in red blood cells, and elevated serum levels of PGI can be a prognostic biomarker for several types of cancers. The PGI activity assay kit is based on the measurement of G6P formed from F6P, and the measurement is coupled to the reduction of the tetrazolium salt INT in a NADPH-coupled enzymatic reaction. Since the INT reaction product exhibits an absorption maximum at 492 nm, the intensity of the red color formed is increased in response to increased PGI activity. The PGI Assay Solution should be stored at -80ºC, and will remain stable for many years if handled properly.
